Overview

Streptococcus pneumoniae capsular polysaccharides are complex carbohydrate polymers that form the outermost layer of the pneumococcal cell wall. These polysaccharides are the primary virulence factors of the bacterium, protecting it from host phagocytosis by masking underlying surface antigens and inhibiting complement deposition [1]. There are over 100 known serotypes, each defined by a unique polysaccharide structure. In the context of pharmacology, these polysaccharides serve as the antigenic components of pneumococcal vaccines, such as the 20-valent pneumococcal conjugate vaccine (PCV20). When conjugated to a carrier protein (like CRM197), they induce a T-cell dependent immune response, leading to high-affinity antibodies and immunological memory [2]. This protection is crucial for preventing invasive pneumococcal diseases, including pneumonia, meningitis, and bacteremia, particularly in vulnerable populations like children and the elderly [3, 4].
