Overview

Streptococcus pneumoniae capsular polysaccharide serotype 10A is a complex surface carbohydrate that serves as a critical virulence factor, shielding the bacterium from host immune recognition and phagocytic clearance. It is composed of repeating oligosaccharide units containing galactose, ribitol-5-phosphate, and glucose, which define its unique antigenic signature. Serotype 10A has been identified as a significant cause of invasive pneumococcal disease, including pneumonia, meningitis, and bacteremia, especially as a 'replacement serotype' following the widespread use of earlier lower-valency conjugate vaccines. To combat this, the polysaccharide is used as a therapeutic target and active component in advanced vaccines like the 20-valent pneumococcal conjugate vaccine (PCV20) and the 23-valent polysaccharide vaccine (PPSV23). These vaccines stimulate the immune system to produce high-affinity, serotype-specific IgG antibodies that facilitate bacterial opsonization and killing by host immune cells.
