Streptococcus pneumoniae capsular polysaccharide serotype 12F is a complex carbohydrate structure that encapsulates the 12F strain of the pneumococcus bacterium, serving as its primary virulence factor [7, 19]. It functions by providing a physical shield that inhibits host immune recognition and prevents opsonophagocytosis by neutrophils and other phagocytes [10, 16]. The chemical structure consists of a hexasaccharide repeat unit containing glucose, galactose, and specialized amino sugars, which can exhibit heterogeneity such as the presence of keto sugar substitutions [1]. Serotype 12F is clinically significant due to its association with outbreaks of invasive pneumococcal disease (IPD), including pneumonia, meningitis, and bacteremia, particularly in populations where earlier conjugate vaccines have led to serotype replacement [2, 5, 8]. As a vaccine target, it is included in multi-valent formulations like the 20-valent pneumococcal conjugate vaccine (PCV20) and the 23-valent pneumococcal polysaccharide vaccine (PPSV23) [3, 13]. These vaccines work by eliciting serotype-specific antibodies that neutralize the protective effect of the capsule, thereby allowing the host's immune system to effectively clear the infection [1, 19].
