Streptococcus pneumoniae capsular polysaccharide serotype 15B is a complex carbohydrate polymer that forms the outermost protective layer of the 15B strain of pneumococcus. This polysaccharide serves as a critical virulence factor by providing an antiphagocytic shield, which allows the bacterium to evade the host's innate immune system and facilitate dissemination into the bloodstream and central nervous system (CDC, 2024). Clinically, serotype 15B is a significant cause of invasive pneumococcal diseases, including bacteremia and meningitis, particularly following the introduction of earlier, lower-valent vaccines that did not cover this serotype. In the pharmaceutical industry, it is utilized as a primary target antigen in multi-valent vaccines such as PCV20 (Prevnar 20) and PCV21 (Capvaxive). These vaccines are designed to elicit opsonizing antibodies that bind to the polysaccharide, enabling phagocytic cells to recognize and destroy the pathogen (Pfizer, 2021; Merck, 2024). The inclusion of 15B in modern formulations is a strategic response to the shifting epidemiology of pneumococcal disease and the need for broader coverage against prevalent serotypes.
As a vaccine antigen, it induces a humoral immune response characterized by the production of serotype-specific opsonophagocytic IgG antibodies. When conjugated to a carrier protein, it triggers a T-cell dependent response that leads to long-term immunological memory (FDA, 2021; Merck, 2024).
