Overview

Streptococcus pneumoniae capsular polysaccharides–diphtheria CRM197 protein conjugates are the active components in several pneumococcal conjugate vaccines. These vaccines consist of purified capsular polysaccharides from specific serotypes of Streptococcus pneumoniae, each chemically linked (conjugated) to the carrier protein CRM197, a non-toxic mutant of diphtheria toxin. The purpose of conjugating S. pneumoniae polysaccharides to CRM197 is to enhance immunogenicity—especially in infants and young children who respond poorly to plain polysaccharide antigens. Upon administration, these glycoconjugates induce T-cell dependent immune responses leading to robust production of anti-capsular antibodies and long-term immunological memory. CRM197 is a genetically detoxified variant of diphtheria toxin produced by Corynebacterium diphtheriae, differing from wild-type by a single amino acid substitution rendering it non-toxic while retaining immunogenicity.
