Streptococcus pneumoniae virulence factors are a diverse array of molecules that enable the bacterium to colonize the host, evade immune detection, and cause systemic disease. The most critical factor is the polysaccharide capsule, which inhibits opsonophagocytosis and defines the various serotypes of the species (Kadioglu et al., 2008). Other significant factors include pneumolysin, a potent toxin that creates pores in host cell membranes, and surface proteins like PspA and PspC that assist in mucosal attachment and complement inhibition (Mitchell & Mitchell, 2010). Unlike traditional drug targets that are inhibited by small molecules, these factors serve as antigens in pneumococcal vaccines such as PCV13 and PPSV23 (CDC, 2023). Vaccination introduces these antigens to the host immune system to stimulate the production of protective antibodies and memory cells. These antibodies subsequently recognize the virulence factors on invading bacteria, facilitating their clearance through the immune system's natural mechanisms such as opsonization and complement-mediated lysis (Paton, 1996).
Vaccines containing these virulence factors act as antigens to stimulate the production of specific antibodies (IgG) and memory B cells. These antibodies then recognize the virulence factors on live bacteria, leading to opsonization, complement activation, and phagocytosis (CDC, 2023).
