Overview

Streptococcus pyogenes cell envelope protease (Spy-CEP), also known as ScpC, is a 170-kDa subtilisin-like serine protease anchored to the cell wall of Group A Streptococcus (GAS) (UniProt P0C0I4). Its primary biological function is the proteolytic cleavage and inactivation of CXC chemokines, most notably interleukin-8 (IL-8/CXCL8), which is a critical chemoattractant for neutrophil recruitment (Edwards et al., 2005, PubMed: 15939750). By degrading these signaling molecules, Spy-CEP allows the bacteria to evade the host's innate immune response, facilitating deep tissue invasion and systemic spread (Sumby et al., 2005, PubMed: 15833928). This enzyme plays a pivotal role in the pathogenesis of severe invasive infections such as necrotizing fasciitis and streptococcal toxic shock syndrome, where a characteristic lack of neutrophils is observed at the infection site (Turner et al., 2009, PubMed: 19126567). Because of its essential role in immune evasion, Spy-CEP is a high-priority target for the development of vaccines and therapeutic inhibitors designed to restore neutrophil-mediated bacterial clearance (Zinkernagel et al., 2008, PubMed: 18337595).

Mechanism of action

Spy-CEP proteolytically cleaves the C-terminal alpha-helix of CXC chemokines, such as Interleukin-8 (IL-8/CXCL8), which is required for their interaction with CXCR1 and CXCR2 receptors on neutrophils, thereby preventing neutrophil recruitment to the site of infection (Zinkernagel et al., 2008, PubMed: 18337595).
