Overview

Streptococcus sanguinis glucosyltransferase (GtfP) is an extracellular enzyme produced by the commensal bacterium S. sanguinis, a primary colonizer of the human oral cavity. This enzyme plays a critical role in the synthesis of alpha-glucans from sucrose, which serve as a sticky extracellular matrix facilitating bacterial adhesion to the tooth surface and the formation of dental plaque biofilms. While S. sanguinis is generally considered a beneficial member of the oral microbiome that competes with cariogenic species like S. mutans, it is also a leading cause of subacute infective endocarditis when it enters the bloodstream. In the context of endocarditis, the glucans produced by GtfP are thought to mediate bacterial attachment to damaged heart valves and fibrin-platelet vegetations. Consequently, GtfP is a significant therapeutic target for anti-virulence strategies aimed at preventing both dental caries and systemic infections. Research into GtfP inhibitors focuses on small molecules and natural products that can disrupt biofilm architecture without exerting broad-spectrum bactericidal pressure, thereby maintaining microbial ecology while reducing pathogenic potential.
