Overview

The stress‑activated protein kinase (SAPK) pathway refers primarily to two subfamilies within the mitogen‑activated protein kinase (MAPK) superfamily: c‑Jun N‑terminal kinases (JNK1/2/3) and p38 MAPKs (p38α/β/γ/δ). These enzymes are activated by various environmental stresses—including inflammatory cytokines, ultraviolet irradiation, heat shock, osmotic changes—and mediate cellular adaptation through phosphorylation cascades that regulate gene expression via transcription factors like c-Jun and ATF2. The SAPK pathways play critical roles in controlling apoptosis, inflammation, cell proliferation/differentiation, and tissue remodeling under pathological conditions such as cancer progression and cardiovascular injury. They have been validated as therapeutic targets; several small-molecule inhibitors have entered clinical trials for inflammatory diseases and cancer but face challenges related to specificity and safety due to their central role in essential cellular processes.
