Overview

Striatin-3 (STRN3) is a calmodulin-binding scaffolding protein that serves as a core regulator within the STRIPAK complex, a multi-protein assembly that critically controls protein (de)phosphorylation and modulates several signaling pathways, including Hippo, MAPK, and cytoskeletal organization[1][3][7]. As a regulatory subunit of protein phosphatase 2A (PP2A), STRN3 helps coordinate kinase and phosphatase activities, influencing cell growth, invasion, and transcriptional regulation[3][1]. STRN3 plays important roles in balancing cell proliferation, migration, and tissue invasion, particularly through cooperative interactions with kinases such as MAP4K4 and via negative regulation of Hippo signaling[1][3][4]. Disruption of STRN3 can impair tumor cell growth and invasion, highlighting its relevance in cancer biology, including reported involvement in hepatocellular carcinoma and medulloblastoma[1][4]. STRN3 is encoded by the STRN3 gene and is mainly localized in the nucleoplasm, Golgi apparatus, and plasma membrane[3].
