Overview

Stromal interaction molecule 1 (STIM1) is a single-pass transmembrane protein residing primarily in the endoplasmic reticulum (ER) membrane. It functions as a crucial calcium sensor, detecting decreases in ER calcium concentration and activating store-operated calcium entry (SOCE) by interacting with ORAI channels at the plasma membrane. This process replenishes intracellular calcium stores and triggers downstream signaling pathways involved in gene expression, cell growth, immune responses, and muscle function. Dysregulation of STIM1 is associated with immunodeficiency, myopathies, and cardiac hypertrophy.
