Overview

Succinate dehydrogenase complex assembly factor 3 (SDHAF3) is a mitochondrial protein that acts as an essential assembly factor for succinate dehydrogenase (also known as complex II of the mitochondrial electron transport chain). It is required for the proper assembly of complex II by promoting the maturation and protection of the SDHB subunit, specifically protecting it from oxidative damage. SDHAF3 is localized to the mitochondria and is involved in cellular energy metabolism by facilitating the function of the electron transport chain and the tricarboxylic acid (TCA) cycle. Its deficiency has been linked to mitochondrial respiratory disorders and possibly some cancers, though it is not itself a direct therapeutic target[1][5]. None currently reported specifically for SDHAF3. Dysfunction in SDHAF3 could (by analogy) result in impaired SDH complex function, which is sometimes monitored by metabolic markers (succinate accumulation), but SDHAF3-specific biomarkers are not established[1].
