Overview

Succinyl-CoA is a critical metabolic intermediate consisting of a succinyl group attached to Coenzyme A via a high-energy thioester bond. It plays a central role in the Citric Acid Cycle (TCA cycle), where it is formed from alpha-ketoglutarate by the alpha-ketoglutarate dehydrogenase complex and subsequently converted to succinate by succinyl-CoA synthetase, generating GTP or ATP in the process [1, 2]. Beyond energy production, Succinyl-CoA is a vital precursor for heme biosynthesis, combining with glycine to form delta-aminolevulinic acid in the mitochondria [3]. It also serves as a substrate for protein succinylation, a post-translational modification that regulates the function of various mitochondrial and cytosolic proteins, thereby influencing metabolic flux [4]. While not a traditional drug target itself, dysregulation of Succinyl-CoA metabolism is linked to several metabolic disorders, including methylmalonic acidemia and succinyl-CoA synthetase deficiency [5]. In oncology, Succinyl-CoA levels are often altered due to mutations in TCA cycle enzymes, contributing to the metabolic reprogramming of cancer cells [6]. Therapeutic strategies typically focus on the enzymes that synthesize or utilize Succinyl-CoA rather than the molecule itself [7].
