Overview

Sulfatase modifying factor 2 (SUMF2) is a protein encoded in humans by the SUMF2 gene. SUMF2 is a member of the sulfatase-modifying factor family and encodes a protein localized to the endoplasmic reticulum lumen, characterized by a DUF323 domain. Although highly homologous to sulfatase modifying factor 1 (SUMF1), SUMF2 lacks catalytic formylglycine-generating activity and is unable to convert newly synthesized inactive sulfatases into their active forms. Instead, SUMF2 is thought to act as a regulatory protein: it heterodimerizes with SUMF1 and can inhibit SUMF1’s activation of sulfatases, suggesting a modulatory role in sulfatase enzyme activity and the regulation of post-translational modifications essential for sulfatase function[1][2][3][4][5][6]. SUMF2’s physiological relevance is demonstrated by its widespread tissue expression and its proposed involvement in the regulation of sulphatase activity, which is critical for cellular metabolism of sulfated molecules. SUMF2 does not have established direct disease-causing mutations, but altered regulation of sulfatases via SUMF2 may influence diseases linked to sulfatase deficiency[3].
