A sulfated glycosaminoglycan is a highly heterogeneous linear polysaccharide that contains sulfated disaccharide repeating units and is mainly found at the cell surface or in the extracellular matrix as part of proteoglycans[1][2]. Major subtypes include heparan sulfate, chondroitin sulfate, dermatan sulfate, and keratan sulfate, all of which are covalently linked to core proteins to form proteoglycans[2]. The sulfation pattern of these chains confers a highly negative charge that determines their ability to interact with diverse proteins, including growth factors, cytokines, morphogens, enzymes, extracellular matrix proteins, and viral pathogens[1][2][3][4]. These interactions make sulfated glycosaminoglycans key modulators of cell adhesion, proliferation, migration, development, and immune responses, as well as common attachment factors or coreceptors for pathogen entry[4][2]. Dysregulation or modification of these molecules, or their engagement by therapeutic agents, can have substantial pathophysiological consequences, including effects on infection, inflammation, cancer, and hemostasis[2][1][4].
