Sulfhydryl groups (–SH), present in cysteine residues of lens proteins (primarily crystallins), are crucial for maintaining lens transparency and structural integrity. They participate in disulfide bond formation and reduction, contributing to protein stability and preventing aggregation. Oxidation of sulfhydryl groups leads to increased disulfide crosslinking, protein insolubility, light scattering, and ultimately, cataract formation. Experimental targeting using sulfhydryl-specific reagents helps study protein conformation and interactions. Free sulfhydryls also act as antioxidants, protecting lens proteins from oxidative damage.
