Overview

Sulfonylurea receptor 1 (SUR1) is a regulatory subunit of ATP-sensitive potassium (K_ATP) channels and certain non-selective cation channels. It is a member of the ATP-binding cassette (ABC) transporter family, encoded by the *ABCC8* gene. SUR1 plays a critical role in linking cellular metabolic state to membrane excitability, particularly in pancreatic beta cells and the central nervous system. It is the molecular target for sulfonylurea class antidiabetic drugs.

Mechanism of action

Sulfonylureas bind to SUR1, inhibiting K_ATP channel activity and leading to membrane depolarization and increased insulin secretion in pancreatic beta cells. In the CNS, SUR1 blockade can prevent brain swelling and enhance neuroprotection following injury.
