Overview

SUMO-activating enzyme subunit 1 (SAE1) is the regulatory subunit of the heterodimeric E1 enzyme complex responsible for activating SUMO (Small Ubiquitin-like Modifier) proteins, including SUMO1, SUMO2, SUMO3, and probably SUMO4[4][7]. SAE1 mediates ATP-dependent activation of SUMO, forming a heterodimer with SAE2 (UBA2), and catalyzes the initial step of the SUMOylation cascade—an essential post-translational modification that regulates protein localization, stability, and function[1][2][4][7]. SAE1 activity is crucial for processes such as chromosome segregation, apoptosis, and cellular stress responses. Dysregulation of SUMOylation and SAE1 expression is implicated in multiple pathologies, especially various cancers and inflammatory conditions[1][6]. SAE1 has potential as a therapeutic target and a biomarker in oncology and immunology[6].
