Superoxide anion (O2•−) is the primary reactive oxygen species (ROS) produced by the partial reduction of molecular oxygen, primarily through the mitochondrial electron transport chain and enzymes like NADPH oxidase. While it serves essential roles in cellular signaling and the immune system's oxidative burst to kill pathogens, an imbalance between ROS production and antioxidant defense leads to oxidative stress. This state causes oxidative damage to lipids, proteins, and DNA, contributing to the pathogenesis of numerous conditions including cardiovascular diseases, neurodegeneration, and cancer. Therapeutic strategies targeting superoxide and its derivatives focus on neutralizing these species to prevent tissue damage. This is achieved through the use of small-molecule scavengers, superoxide dismutase (SOD) mimetics, or drugs that enhance endogenous antioxidant capacity. However, because ROS also function as critical secondary messengers in pathways like cell proliferation and vasodilation, non-selective depletion can lead to adverse effects. Modern drug development aims for site-specific or mitochondria-targeted antioxidants to mitigate these risks while addressing the underlying oxidative pathology.
