Overview

Suppressor APC domain containing 2 encodes a protein of approximately 42.3 kDa, initially recognized as a cell cycle-associated molecule critical for mitotic progression. SAPCD2 is not expressed in most healthy postnatal tissues, but its expression is strongly upregulated during embryogenesis and in various tumors. Structurally, SAPCD2 features a coiled-coil domain and proline-rich regions facilitating protein-protein interactions. Its cellular role encompasses regulation of spindle orientation, symmetric cell division, and modulation of the Wnt/β-catenin pathway—where SAPCD2 competes with protein phosphatase 2A (PP2A) for Axin1 binding, thereby reinforcing β-catenin phosphorylation and suppressing Wnt target gene activity[1]. SAPCD2 is being studied as both a diagnostic and therapeutic cancer target, and its interactions with key cell division and signaling proteins are partly understood but remain the subject of ongoing research. Recent evidence suggests it may play additional roles in inflammation and developmental biology[1].
