Suppressor of cytokine signaling 2 is a cytoplasmic adapter protein containing a central SH2 domain and a C-terminal SOCS box. It is upregulated by cytokine stimulation and negatively regulates JAK-STAT signaling either by direct inhibition or by kinase receptor degradation via ubiquitination. SOCS2 is a key feedback inhibitor controlling growth hormone/IGF-1 signaling, proliferation, and immune cell differentiation, particularly in T cells and NK cells. Pathologic perturbation of SOCS2 function can contribute to abnormal growth, cancer, immune dysfunction, and metabolic diseases. Key features include: Negative regulator of cytokine and growth hormone signaling; Interacts with insulin-like growth factor 1 receptor, erythropoietin receptor, leptin receptor, and components of the JAK-STAT pathway; Functions as substrate recognition subunit in a cullin5 E3 ubiquitin ligase complex; No currently approved direct-targeting drugs, but significant interest as a mediator for targeted protein degradation strategies.
