Overview

Suppressor of cytokine signaling 4 (SOCS4) is an intracellular protein belonging to the SOCS family, functioning as a negative regulator of cytokine and growth factor signaling[1][2][4][5]. It contains a central SH2 domain and a C-terminal SOCS box that mediates its role in targeting signaling proteins for ubiquitination and proteasomal degradation[1][2][3]. SOCS4 is distinguished within its family by a long N-terminal domain, and although its exact physiological functions are still being elucidated, evidence indicates its involvement in tuning cytokine signaling in immune cells and potentially regulating signaling by receptor tyrosine kinases like EGFR[4]. Mouse models indicate that SOCS4 is important in anti-viral immune responses, particularly in T cell activation and the clearance of influenza virus through modulation of cytokine production, chemokine signaling, and CD8+ T cell trafficking; its absence can lead to increased susceptibility to infection and dysregulated inflammation[4]. There is limited information on direct roles in human disease, and it is not currently a druggable target.

Mechanism of action
