Overview

Suppressor of cytokine signaling 6 (SOCS6) is an intracellular protein encoded by the SOCS6 gene and a member of the SOCS family, which functions as a negative feedback regulator of cytokine receptor signaling pathways[4][5][6]. SOCS6 is a multi-domain protein with an SH2 domain and a SOCS box, enabling it to interact with phosphorylated tyrosine residues and participate as part of an E3 ubiquitin-protein ligase complex, leading to ubiquitination and subsequent proteasomal degradation of target proteins[5]. Unlike most SOCS proteins, SOCS6 predominantly modulates receptor tyrosine kinase signaling rather than classical cytokine receptor/JAK-STAT signaling and has additional roles in mitochondrial dynamics and apoptosis[1][2]. SOCS6 gene loss or reduction is observed in multiple cancers and is associated with increased cell survival and impaired apoptosis[2]. The protein is not a receptor, enzyme, transporter, or common direct drug target, but is considered for its regulatory role in cancer and immune signaling[1][2][5].
