Overview

Suppressor of cytokine signaling 6 (SOCS6) is a member of the SOCS family of proteins that act as negative regulators of cytokine and growth factor signaling (UniProt: O14544). It contains a central SH2 domain and a C-terminal SOCS box, which allows it to function as a substrate-recognition component of Cullin-RING E3 ubiquitin ligase complexes (PubMed: 12750429). SOCS6 specifically targets the insulin receptor substrates (IRS1 and IRS2) and the KIT receptor tyrosine kinase for ubiquitination and subsequent proteasomal degradation, thereby modulating insulin sensitivity and cellular proliferation (PubMed: 15611074). In many clinical contexts, SOCS6 is recognized as a tumor suppressor, and its downregulation—often mediated by microRNAs like miR-155 or promoter hypermethylation—is associated with the progression of various cancers, including gastric and lung carcinomas (PubMed: 25670453). While no small-molecule drugs are currently approved for SOCS6, it is a significant focus of research involving microRNA-based therapeutics and strategies to restore its expression in malignant cells. Its dual role in regulating both insulin sensitivity and oncogenic signaling makes it a complex but promising target for metabolic and oncological interventions.
