Overview

Suppressor of cytokine signaling protein 3 (SOCS3) is a protein encoded by the SOCS3 gene and a member of the SOCS family of cytokine-induced inhibitors. SOCS3 contains three modular domains: an N-terminal kinase inhibitory region (KIR), a central Src homology 2 (SH2) domain that binds phosphotyrosine residues on cytokine receptors, and a C-terminal SOCS box that assembles components of an E3 ubiquitin ligase complex. SOCS3 functions as a key negative regulator of cytokine signal transduction, particularly of the JAK-STAT pathway, by directly inhibiting the catalytic activity of Janus kinases (JAK1, JAK2, TYK2) and promoting degradation of activated receptors. Its biological effects span immune regulation, inflammation, cell proliferation, and metabolic signaling. SOCS3 is implicated in several diseases, including cancer, inflammatory conditions, insulin resistance, and possibly neurodegenerative disorders. Although SOCS3 itself is not yet a therapeutic target for approved drugs, it is central to the mechanism of action of many drugs targeting the JAK-STAT pathway, and its expression serves as a potential biomarker for immune and metabolic dysfunction
