Overview

Suppressor of fused homolog (SUFU) is a protein that acts as a central negative regulator within the Hedgehog signaling pathway by binding and inhibiting the activity of Gli transcription factors, preventing their nuclear accumulation and transcriptional activation of Hedgehog target genes. SUFU's regulatory influence is crucial for normal embryonic development and the suppression of inappropriate cell proliferation; mutations or loss of SUFU function are linked to cancer predilection (notably medulloblastoma and basal cell carcinomas) and disorders of embryogenesis. SUFU exerts its tumor suppressor activity by tethering Gli proteins in the cytoplasm, promoting repressor forms of Gli, and recruiting transcriptional co-repressors in the nucleus. While SUFU itself is not currently a direct drug target, modulation of the HH pathway through upstream effectors indirectly relies on SUFU regulation for their therapeutic efficacy.
