Suppressor of IKBKE 1 (SIKE1) is a physiological suppressor protein that inhibits the activity of IKK-epsilon (IKBKE) and TBK1, kinases involved in the activation of IRF3—a transcription factor necessary for type I interferon production[1][3][4][5][6][8]. SIKE1 acts by disrupting interactions between IKBKE or TBK1 and proteins in the virus- and TLR3-triggered IRF3 activation pathway, thereby selectively downregulating interferon-stimulated gene expression without affecting NF-kappa B signaling[1][3][4][8]. It is a component of the striatin-interacting phosphatase and kinase (STRIPAK) complex, serving as a bridging scaffold and regulating the Hippo signaling pathway, which controls cell growth and organ size[2][1]. SIKE1 is widely expressed in human tissues, and while changes in its function may influence infection susceptibility or cell proliferation, it is not an established therapeutic target and no drugs are known to act directly upon it[1][3][9].
