Supragingival calculus, commonly known as dental tartar, is a mineralized deposit of dental plaque that forms on the clinical crowns of teeth above the gingival margin (StatPearls, 2023). It is primarily composed of an inorganic crystalline matrix—including hydroxyapatite, whitlockite, and octacalcium phosphate—interspersed with organic components such as desquamated epithelial cells, leukocytes, and a dense network of mineralized microorganisms (PubMed, PMID: 2824334). While supragingival calculus itself is not the primary etiologic agent of periodontal disease, its porous and rough surface acts as a reservoir for living pathogenic bacteria, which can lead to gingival inflammation and the progression of periodontal conditions (NIH, 2022). Therapeutic intervention is typically mechanical, involving professional scaling and root planing, though chemical agents like pyrophosphates and zinc salts are incorporated into dentifrices to inhibit the crystallization process and prevent new deposits (PubMed, PMID: 15127869). Because it is a physical mineralized mass rather than a specific biological macromolecule like a receptor or enzyme, it is not classified as a traditional therapeutic target for drug discovery, but rather a clinical target for oral hygiene management.
