Surface Layer Protein A (SlpA) is the primary structural component of the S-layer, a paracrystalline protein array forming the outermost surface of certain bacteria like *Clostridioides difficile*. It is crucial for bacterial structure, adherence, and virulence. SlpA undergoes post-translational cleavage into HMW and LMW subunits. It also acts as a receptor for bacteriophages. Sequence variability among strains impacts immune evasion and susceptibility to phage therapy.
