A surfactant protein B mimic is a synthetic peptide or protein analog designed to replicate the essential biophysical and functional characteristics of native surfactant protein B (SP-B), which is a small, amphipathic, saposin-like protein crucial for the formation and stability of pulmonary surfactant in the lung alveoli[1][2][3][5]. These mimics are engineered to retain the key regions and structural motifs of SP-B (such as the N- and C-terminal α-helices, amphipathic surfaces, and disulfide-linked topology) critical for interaction with phospholipids and lowering of surface tension at the air-liquid interface in the lungs. SP-B mimics, including peptides like Mini-B, Super Mini-B, and B-YL, are used as active agents in synthetic surfactant formulations for treating respiratory distress conditions, particularly neonatal respiratory distress syndrome (RDS) and acute respiratory distress syndrome (ARDS), where natural surfactant is deficient or dysfunctional[1][2][3][5]. Synthetic lung surfactants with these mimics represent important therapeutic tools, especially where animal-derived proteins are impractical, unavailable, or pose immunogenicity risks.
