The Survivin peptide-MHC class I complex is formed when a short peptide derived from the survivin protein—a key inhibitor of apoptosis implicated in cancer—is bound within the groove of an MHC class I molecule, typically on the surface of cancer cells. This complex is presented to cytotoxic T cells (via their T-cell receptors), facilitating immune recognition and targeted cell destruction. Therapeutic strategies exploit this complex to enhance anti-tumor immunity, notably through peptide vaccines and engineered T-cell therapies. Successful immune targeting depends on the stability and affinity of peptide binding to the MHC molecule, which can be optimized by modifying peptide anchor residues. Other safety and efficacy factors include HLA allele selectivity and tumor immune evasion mechanisms such as loss or dysfunction of MHC class I surface expression. This molecular complex is under active investigation as a cancer immunotherapy target and biomarker.
T-cell activation: cytotoxic T lymphocytes (CTLs) recognize survivin peptide displayed by MHC class I, leading to lysis of cancer cells presenting this complex. Immune modulation: peptide modifications (e.g., anchor residue substitutions) can increase CTL activation by improving peptide binding to MHC I.
