Overview

Sushi domain-containing protein 3 (SUSD3) is a membrane-associated protein containing a Sushi domain, involved in cell adhesion, proliferation, and migration, especially within the context of estrogen receptor–positive breast cancer[1][2][3][5]. SUSD3 is upregulated by estradiol via direct ERα recruitment, localizes to the plasma membrane, and contributes to actin cytoskeletal remodeling, thereby facilitating cell movement and tumor progression[1][3]. It has been identified as a potential biomarker for endocrine therapy sensitivity, notably aromatase inhibitors, and its expression is associated with improved responsiveness to therapy in ER-positive tumors[1][2][3][5]. Recent studies also demonstrate a pivotal role in modulating immune cell activity and infiltration, positioning SUSD3 as a promising therapeutic target across multiple cancer types and as a prognostic marker for immunotherapy outcomes[2][5]. There are currently no approved drugs directly binding or inhibiting SUSD3, but computational screens and molecular docking suggest it may be sensitive to certain anticancer agents[2][5].
