Overview

SVIL antisense RNA 1 is a long noncoding antisense RNA located on chromosome 10q13. It is downregulated in lung adenocarcinoma tissues and correlates with better patient prognosis. Functionally, SVIL-AS1 restrains proliferation and promotes cell cycle arrest in lung adenocarcinoma cells by facilitating the proteasomal degradation of E2F1, a key cell cycle regulator. The RNA methyltransferase METTL3 installs m6A methylation marks on SVIL-AS1, increasing its stability and enhancing its tumor-suppressive activity. The mechanism links m6A-modified lncRNA function with regulation of cell cycle and cancer progression.

Mechanism of action

For experimental manipulations, SVIL-AS1 acts by increasing its expression (via METTL3-mediated m6A RNA methylation) to suppress proliferation and promote cell cycle arrest, mediated through degradation of E2F1 and downregulation of its target genes
