SWELL1, also known as LRRC8A, is a protein that forms the core of the volume-regulated anion channel (VRAC). VRACs are critical for maintaining cellular homeostasis by regulating cell volume in response to osmotic changes. SWELL1 can form homo-hexameric channels but typically assembles with other LRRC8 family members (LRRC8B-E) to create heteromeric VRAC complexes, which dictates the biophysical properties and selectivity of the channel. It plays a role in metabolic regulation, cell survival under osmotic stress, and is associated with diseases such as metabolic syndrome and immunodeficiency.
