Switch-associated protein 70 (SWAP70) (SWAP70)

SWAP70 (Switch-associated protein 70) is a multifunctional protein that acts as a guanine nucleotide exchange factor (GEF) for Rac GTPases and an actin-binding protein, playing a critical role in B-cell signaling and cytoskeletal reorganization (UniProt Q9UH65). It is uniquely characterized by its ability to bind phosphatidylinositol 3,4,5-trisphosphate (PIP3) via its Pleckstrin Homology (PH) domain, which facilitates its translocation to the plasma membrane upon cell activation (NCBI Gene ID: 23075). In the immune system, SWAP70 is essential for B-cell maturation, migration, and immunoglobulin class switching (PubMed: 10675333). In the context of disease, SWAP70 is frequently overexpressed in various malignancies, including glioblastoma and breast cancer, where it promotes tumor cell migration, invasion, and metastasis (PubMed: 25605123). While no clinical-stage drugs currently target SWAP70 mRNA or protein directly, experimental approaches using RNA interference (siRNA) have demonstrated potential in reducing tumor progression in preclinical models (PubMed: 30105498). Therapeutic modulation of SWAP70 presents a challenge due to its essential role in normal immune function, necessitating highly specific targeting strategies to avoid systemic immunosuppression.
