Overview

SWAP70 (Switching B cell complex subunit SWAP70) is a phosphatidylinositol 3,4,5-trisphosphate-dependent guanine nucleotide exchange factor (GEF) that functions as a signaling adapter, primarily expressed in B lymphocytes[3][4]. It plays a central role in transducing signals from cell surface receptors (e.g., the B cell receptor) to intracellular actin cytoskeleton remodeling via the activation of Rac GTPases[1][3][4]. SWAP70 is essential for proper B cell polarization, homing, adhesion, membrane ruffling, and effective migration into secondary lymphoid organs, all of which are critical for normal B cell immune function[2]. It is also dynamically localized between cytoplasm, plasma membrane, and nucleus during the activation and differentiation of B cells, and is directly involved in immunoglobulin class switch recombination and B cell interaction with other immune molecules[4].
