Synapse differentiation-inducing gene protein 1 (SYNDIG1) is a brain-enriched, transmembrane protein belonging to the interferon-induced transmembrane family, highly conserved across vertebrates[3][5]. It plays a critical role in the formation, maturation, and maintenance of excitatory synapses, primarily by regulating the content and activity of AMPA-type glutamate receptors at nascent synapses and contributing to postsynaptic development[1][6]. Alteration of SYNDIG1 expression levels can significantly impact synapse number, size, and stability, thereby influencing neural circuit function and potentially contributing to neurodevelopmental disorders such as epileptic encephalopathies and, more broadly, to learning, memory, and cognitive functions[1][3][6].
