Overview

Synapsin-2 is a neuron-specific phosphoprotein that associates with the cytoplasmic surface of synaptic vesicles and binds to the cytoskeleton in presynaptic nerve terminals[1][2][6]. It is crucial for the regulation of neurotransmitter release and synaptic vesicle trafficking. Through alternative splicing, the SYN2 gene encodes the isoforms synapsin IIa and synapsin IIb, both of which play roles in maintaining synaptic vesicle pools and synaptic plasticity[1]. Synapsin-2 is implicated in normal synaptogenesis and neurotransmission, and its dysfunction is associated with several neuropsychiatric and neurodevelopmental disorders, including schizophrenia, autism, epilepsy, and bipolar disorder[2][3]. There are currently no widely recognized small molecule drugs or direct inhibitors/activators for Synapsin-2, but it is considered a protein of high therapeutic interest, particularly for neurological and psychiatric indications[3].
