C22orf39 (Synaptic plasticity regulator PANTS) is a protein primarily implicated in the regulation of synaptic plasticity, which is crucial for learning and memory processes in the brain. It negatively regulates long-term potentiation, a process that strengthens synaptic connections, and modulates adult synaptic plasticity. The protein influences synaptic signaling dynamics by stabilizing the interaction between RTN4/Nogo-A and its receptors, thereby preventing clustering of postsynaptic AMPA receptors. Upon neuronal stimulation, C22orf39 is degraded at synapses, facilitating AMPA receptor clustering involved in synaptic adaptation. While associated with certain immune cell deficiencies and chromosomal variation syndromes, C22orf39 is not currently recognized as a classic therapeutic target and its clinical implications are still under investigation.
