Synaptogenesis is the complex biological process by which neurons form new functional connections, known as synapses, within the nervous system (NIH, 2023). This process is most prolific during embryonic and early postnatal development but remains a critical mechanism for synaptic plasticity, learning, and memory throughout adulthood (PubMed, 2021). It involves the coordinated recruitment of presynaptic vesicles and the assembly of postsynaptic receptors, facilitated by cell adhesion molecules like neuroligins and neurexins (StatPearls, 2023). Dysregulation or failure of synaptogenesis is a primary pathological feature in neurodevelopmental disorders like autism and schizophrenia, while the progressive loss of synapses is the most significant correlate of cognitive decline in neurodegenerative diseases such as Alzheimer's disease (NIH, 2022). Although synaptogenesis is a physiological process rather than a single molecular target, it is a major therapeutic objective in drug discovery; compounds like ketamine and neurotrophic factors are frequently evaluated for their ability to promote synaptic density and restore functional neural circuits (PubMed, 2020).
Modulation of synaptogenesis involves the induction of dendritic spine growth, recruitment of synaptic proteins (e.g., PSD-95, Synaptophysin), and activation of neurotrophic signaling pathways such as the BDNF/TrkB axis.
