Synaptogyrin-4 is an integral membrane protein and one of four synaptogyrin family members, each of which contains four transmembrane domains. The synaptogyrin family is known for their involvement in vesicle membrane trafficking and structural organization, particularly in neural tissues. While other synaptogyrins (such as synaptogyrin-1 and -3) are highly expressed in neurons and are key components of synaptic vesicles for neurotransmitter release, synaptogyrin-4 is the least characterized member. It does not co-localize with classic synaptic vesicle markers in neurons. The literature suggests synaptogyrin-4 may be involved in promoting internalization and retrograde trafficking of vesicle populations, potentially inhibiting exocytic release of protein cargo—drawing an analogy to related proteins like synaptogyrin-2 in non-neuronal cells. Recent findings indicate synaptogyrin-4 is upregulated in motor neurons during the transition from presymptomatic to symptomatic phases in ALS mouse models, suggesting a possible link to neurodegenerative disease pathophysiology. One published protein-protein interaction identifies the glucagon-like peptide-1 receptor (GLP-1R) as a possible SYNGR4 binding partner, though independent validation is lacking. There are currently no known drugs or small molecules that modulate synaptogyrin-4, nor is there evidence it is directly implicated as a therapeutic target in any current clinical or investigative context. Its primary significance is in basic neuroscience research and understanding vesicular biology rather than direct therapeutic relevance.
