Overview

Synaptonemal complex protein 2 (SYCP2) is an essential meiosis-specific structural protein that forms a critical component of the axial/lateral elements of the synaptonemal complex—a scaffold that physically links homologous chromosomes during prophase I of meiosis[1][2][4]. SYCP2 is required for correct assembly and function of the synaptonemal complex, ensuring homologous chromosome synapsis, pairing, and recombination[1][2][3][4]. Through direct interactions with other synaptonemal complex proteins (e.g., SYCP1, SYCP3), SYCP2 is necessary for the integration and organization of chromatin during gametogenesis. Disruption of SYCP2, whether by loss-of-function mutations or aberrant regulation, impairs complex assembly and leads to defective spermatogenesis, resulting in male infertility. Ectopic expression of SYCP2 has also been detected in certain premalignant tissues, implicating its dysregulation in genomic instability and potentially linking it to some cancers[1].
