Synaptophysin-like 2 (SYPL2), also known as Mitsugumin-29 (MG29), is a transmembrane protein essential for the structural and functional integrity of skeletal muscle (UniProt Q9HCG1). It belongs to the synaptophysin family and is characterized by the presence of a MARVEL domain, which facilitates membrane apposition and vesicle trafficking (PubMed: 10364154). SYPL2 is localized to the triad junctions, where it plays a pivotal role in excitation-contraction (E-C) coupling by maintaining the proximity between the transverse tubules and the sarcoplasmic reticulum (PubMed: 16707444). Research using knockout models has demonstrated that a lack of SYPL2 results in fragmented triad structures, altered calcium signaling, and increased susceptibility to muscle fatigue (PubMed: 10364154). Furthermore, the expression of SYPL2 is known to decrease with age, contributing to the progression of sarcopenia and muscle atrophy (PubMed: 16707444). While SYPL2 is a critical component of muscle physiology and a potential target for treating age-related muscle decline, there are currently no approved drugs or clinical-stage compounds that specifically modulate its activity.
