Overview

SNAP-25 (Synaptosomal-associated protein 25 kDa) and synaptobrevin (VAMP1/VAMP2) are core components of the SNARE complex, a molecular machine essential for calcium-triggered synaptic vesicle fusion and neurotransmitter release at the neuromuscular junction and cholinergic nerve terminals[2][3][4][5][6]. Cleavage of either protein by specific neurotoxins (notably botulinum and tetanus toxins) prevents SNARE complex assembly, thereby abolishing acetylcholine exocytosis and inducing rapid, profound paralysis. These proteins are integral to normal synaptic transmission, and their disruption underlies the mechanism of action for various neurotoxins and therapeutic agents targeting neurotransmitter release.
