Overview

Synaptotagmin 6 is a member of the synaptotagmin family of membrane-trafficking proteins, characterized by an N-terminal transmembrane region and two C-terminal C2 domains (C2A and C2B) capable of binding calcium[1][3]. It functions as a calcium sensor involved in the regulation of neurotransmitter release and hormone secretion, playing a key role in calcium-dependent exocytosis in neurons and endocrine cells[1][3]. Among at least 17 mammalian synaptotagmins, Synaptotagmin 6 is one of eight isoforms with calcium-binding activity, underpinning its role in synchronizing and regulating fast synaptic transmission through vesicle fusion at the presynaptic membrane[1]. While closely related to other synaptotagmins (such as SYT1, SYT2, and SYT7), SYT6 is distinct in its expression profile and specific biochemical properties. Familial dysfunction or depletion of synaptotagmins has been linked to neuropsychiatric disorders such as bipolar disorder and may participate in neurodegenerative processes and injury response[1]. However, the druggability of SYT6 remains largely unexplored, and no approved pharmaceuticals directly modulate its function at present.
