Synaptotagmin-like protein 4 (SYTL4) is a member of the synaptotagmin-like (Slp) family, characterized by an N-terminal Rab27-binding domain and tandem C2 domains at the C-terminus. Unlike synaptotagmins, SYTL4's C2 domains do not require calcium for membrane binding and participate in calcium-independent vesicle trafficking. SYTL4 functions as an effector for Rab27 GTPases, modulating the trafficking and exocytosis of dense-core granules, thereby regulating hormone secretion in tissues such as the pancreas and pituitary. Mutations or dysregulation of SYTL4 are associated with certain rare diseases and implicated in vesicle transport anomalies, but it is not considered a classical pharmacological target or receptor.
