Overview

Syncytin-2, encoded by the HERV-FRD provirus ancestral env polyprotein gene, is an envelope protein of endogenous retroviral origin expressed in human placenta. Unlike most retroviral envelopes, syncytin-2 mediates cell–cell fusion in the placenta, playing a crucial role in the formation of the syncytiotrophoblast, which is essential for fetomaternal nutrient exchange[2][3]. The gene is uniquely expressed in placental tissue and is highly conserved among simians[2]. Unlike some other HERV env proteins, syncytin-2 does not appear to play a role in viral infection or classical signaling, and is not a receptor, transporter, or classical therapeutic target. No drug interactions or known safety concerns are linked to this gene.

Biological functions

Placenta formation: Induces cell–cell fusion in the developing human placenta[2][3].Syncytiotrophoblast formation: Drives the formation of the multinucleated outer layer of the placenta, essential for maternal-fetal interface[2].

Disease associations

Other (not linked to specific diseases, though research investigates HERVs for possible roles in cancer, autoimmunity, and placental pathologies. No direct, widely accepted causal connection for HERV-FRD/Syncytin-2)[1].
