Overview

The Syndecan-1-derived HLA-A2 peptide is a specific tumor-associated antigen complex consisting of a peptide fragment derived from the Syndecan-1 (CD138) protein presented by the Human Leukocyte Antigen A2 (HLA-A2) MHC class I molecule. Syndecan-1 is a transmembrane heparan sulfate proteoglycan that is highly overexpressed in multiple myeloma and various solid tumors, where it promotes cell proliferation, adhesion, and angiogenesis. In the context of immunotherapy, specific immunogenic peptides from Syndecan-1, such as the GLVGLIFAV sequence, are processed and presented on the cell surface of malignant cells. This peptide-MHC complex serves as a precise target for T-cell receptor (TCR)-engineered T-cell therapies and peptide-based vaccines. By targeting this specific epitope, the immune system can be directed to selectively identify and eliminate cancerous cells while minimizing damage to healthy tissues that do not present the peptide at similar levels.
