Synergin gamma (SYNRG) is an accessory protein that interacts specifically with the gamma subunit of the AP-1 clathrin-adaptor complex, which functions at the trans-Golgi network in the sorting and trafficking of certain membrane and secretory proteins. SYNRG acts as a linker, connecting the AP-1 complex to other molecular components involved in clathrin-coated vesicle formation and membrane trafficking, thus regulating protein sorting and trafficking between the trans-Golgi network and endosomes. Loss of function disrupts trafficking of key cargo proteins and regulated secretion from specialized organelles.
