Synovial sarcoma X breakpoint protein 2 (SSX2) is a member of the SSX family of transcriptional repressors implicated in chromosomal translocations, specifically the t(X;18) translocation found in synovial sarcomas. This translocation results in the fusion of the SYT gene with SSX genes, commonly SSX1 or SSX2, leading to the formation of an oncogenic fusion protein. SSX2 elicits immune responses in cancer patients, making it a potential target for cancer vaccine-based immunotherapy. Mutations such as fusions and other variants are observed in various cancers, including hematopoietic and lymphoid malignancies.
SSX2 is not directly drugged; the fusion protein SYT-SSX is the target of interest, and immunotherapy strategies target aberrant expression patterns.
